Description
Formaldehyde belongs to the carbonyl compounds, a subclass of organooxygen compounds within the organic compounds. It has the chemical formula CH2O and an average molecular weight of 30.03 g/mol. At room temperature, this exogenous compound exists as a liquid. Formaldehyde serves diverse industrial and biological roles. It is used as a biocide, binder, preservative, adhesive (PMC12259876), fungicide (PMC3136160), and reducing agent (PMC7711730, PMC8003255, PMC8426366). Additionally, it functions as a monomer, an electron donor (PMC6648437), an adhesion/cohesion promoter, and an insulator. In biological contexts, it acts as an allergen, a metabolite (PMC7618990), a carcinogenic agent, and an inhibitor of EC 3.5.1.4 (amidase). It is also utilized as a disinfectant. The compound is released from 69 recorded sources, including building and construction materials (roofs, floors, ceilings), food processing, pesticides, and household cleaning products. It is used in Acrylonitrile butadiene styrene and released in Polypropylene, Polyamide, and Polyethylene Terephthalate. Exposure occurs via touch, oral intake, and inhalation (PMC10005944, PMC11938325, PMC13309467, PMC3701998, PMC4097170, PMC4365752), as well as through smoking (PMC11938325). Formaldehyde is detected in numerous tissues, including the brain, liver, lung, kidney, and skin. It targets the D-alanyl-D-alanine carboxypeptidase and Transient receptor potential cation channel subfamily A member 1 (TRPA1), and it forms cross links between protein and DNA. Health effects include cancer (PMC10005944, PMC10218431, PMC12388048, PMC12909993, PMC2891118, PMC3923370, PMC4949043, PMC7610456, PMC8869180, PMC9104621, PMC9925107), leukemia (PMC3852791, PMC4097170, PMC8375462), myeloid leukemia (PMC12259876), and increased lung cancer (PMC3136160). Other associated conditions include asthma (PMC12337040, PMC1241209, PMC7722407), allergic contact dermatitis (PMC1456588, PMC9299984), pulmonary edema (PMC8215519), renal failure (PMC12986066), and increased blood pressure (PMC9498982).

Toxicity Profile
Mechanism of ToxicityIt is likely that formaldehyde toxicity occurs when intracellular levels saturate formaldehyde dehydrogenase activity, allowing the unmetabolized intact molecule to exert its effects. Formaldehyde is known to form cross links between protein and DNA and undergo metabolic incorporation into macromolecules (DNA, RNA, and proteins). (24)
Carcinogenicity (IARC Classification)1, carcinogenic to humans. (23)
Minimum Risk LevelAcute Inhalation: 0.04 ppm (22) Intermediate Inhalation: 0.03 ppm (22) Chronic Inhalation: 0.008 ppm (22) Intermediate Oral: 0.3 mg/kg/day (22) Chronic Oral: 0.2 mg/kg/day (22)
SymptomsLow levels of formaldehyde can cause irritation of the eyes, nose, throat, and skin. (24)
