Description
Bisphenol A (C15H16O2) is an exogenous organic compound with an average molecular weight of 228.29 g/mol. It exists as a solid at room temperature. Bisphenol A belongs to the diphenylmethanes, a subclass of benzene and substituted derivatives within the organic compounds. Industrially, it is used as a catalyst (PMC11381038), additive (PMC13025183), stabilizer (PMC4018667), antioxidant (PMC4018667), and fungicide (PMC5588144), as well as a flame retardant (PMC5588144). It is present in high-density polyethylene and used in polyurethane, polyvinyl chloride, acrylonitrile butadiene styrene, and low-density polyethylene. It is released from polycarbonate, polyamide, polystyrene, polypropylene, and polyethylene terephthalate. Biologically, it acts as an estrogen (PMC5505257), xenoestrogen (PMC11105698), agonist (PMC10253990), inhibitor (PMC4452124), electron donor (PMC10624391), toxin (PMC9457803), and endocrine disruptor (PMC10043879, PMC13347373). It is an environmental contaminant (PMC8130848) and phenol (PMC9518589). It targets several proteins, including the estrogen receptors ESR1 and ESR2, the androgen receptor AR, and the estrogen-related receptor gamma (ESRRG). It also competitively inhibits the binding of L-T3 and 17beta-estradiol to protein disulfide-isomerase (P4HB). Exposure occurs via touch, oral, and inhalation routes (PMC10047534, PMC11546519, PMC3419714, PMC6024920, PMC6098722, PMC6370520, PMC9098901). Sources include drinking water, aquatic systems, food contact materials, building and construction materials, and electrical equipment. Health effects associated with bisphenol A include cancer (PMC10139116, PMC12758521, PMC6625918, PMC7766742, PMC8151977, PMC8395949, PMC8746877, PMC9300791), specifically breast cancer (PMC10314155, PMC11755457, PMC12320938, PMC13064397, PMC8834979, PMC11873108, PMC11938062, PMC12573425, PMC12649852, PMC13179207, PMC5458606, PMC5521805, PMC7064327), colorectal cancer (PMC13064397), endometrial cancer (PMC10720226, PMC6891210), and ovarian cancer (PMC11479995). It is linked to prostate cancer (PMC10938891, PMC11357130, PMC2072856), though some data suggests a decrease (PMC11613181, PMC12573425, PMC3458140). Other associations include obesity (PMC10048338, PMC10226345, PMC10314155, PMC10596897, PMC10831155, PMC10938891, PMC11277516, PMC11443065), infertility (PMC10139116, PMC10612124, PMC10831155, PMC10938891, PMC4562201, PMC8151977), male infertility (PMC10261808, PMC13193906), polycystic ovary syndrome (PMC10938891, PMC11479995), endometriosis (PMC13179207), hypertension (PMC12284880, PMC4985394, PMC7684853, PMC9921712), increased blood pressure (PMC12875064, PMC7354351), stroke (PMC10946040), atherosclerosis (PMC9469194), coronary artery disease (PMC12875064), chronic kidney disease (PMC12829686), inflammation (PMC11319549, PMC6397216), and asthma (PMC12501166, PMC12537159, PMC9918139), though some reports indicate a decrease (PMC12321497, PMC7722407).

Toxicity Profile
Mechanism of ToxicityBisphenol is an endocrine disruptor. Low doses of bisphenol A can mimic the body's own hormones, possibly causing negative health effects. There is thus concern that long term low dose exposure to bisphenol A may induce chronic toxicity in humans (6).
Carcinogenicity (IARC Classification)No indication of carcinogenicity to humans (not listed by IARC).
Minimum Risk LevelNot Available
SymptomsBisphenol A has low acute toxicity, with an oral LD50 of 3250 mg/kg in rats, but it is an endocrine disruptor. Low doses of bisphenol A can mimic the body's own hormones, possibly causing negative health effects. There is thus concern that long term low dose exposure to bisphenol A may induce chronic toxicity in humans. (7)
TreatmentConsider dilution immediately after ingestion. Insertion of a specialized nasogastric tube after confirmation of a circumferential burn may prevent strictures. In case of inhalation, move patient to fresh air and monitor for respiratory distress. If cough or difficulty breathing develops, evaluate for respiratory tract irritation, bronchitis, or pneumonitis. In case of eye contact, irrigate exposed eyes with copious amounts of room temperature water for at least 15 minutes. Decontaminate skin or mucous membranes immediately with water and a soft sponge.
