Description
Potassium nitrate (KNO3) is an inorganic compound with an average molecular weight of 101.10 g/mol. Potassium nitrate belongs to the alkali metal nitrates, a subclass of alkali metal oxoanionic compounds within the inorganic compounds. This exogenous substance is a solid at room temperature and is classified as a mixed metal/non-metal compound. The compound is utilized as a fertilizer, as well as an industrial humectant, biocide, dye, and flavouring and nutrient. It is found in or released from 34 recorded sources, including food, food processing and cookware—specifically the processing of meats and fermentation processes for beer, wine, sparkling wine, and the preparation of malt and vinegar. It is also found in building and construction materials such as floors, tents or canopies; household cleaning and consumer products including bleaching agents, match receptacles or boxes, cigar cigarettes, and other smoking requisites; and electrical and electronic equipment such as amplifiers, antennas, communication cables or conductors, and adjustable resistors. Potassium nitrate is used as a treatment for botulism (PMC8467560) and is associated with colorectal cancer (PMC10301060). Regarding protein targets, it acts as an agonist of the voltage-gated inwardly rectifying potassium channel KCNH2 and interacts with hemoglobin subunit alpha (HBA1) and hemoglobin subunit beta (HBB).

Toxicity / Toxin ClassNot Available
Mechanism of ToxicityNitrate's toxicity is a result of it's conversion to nitrite once in the body. Nitrite causes the autocatalytic oxidation of oxyhemoglobin to hydrogen peroxide and methemoglobin. This elevation of methemoglobin levels is a condition known as methemoglobinemia, and is characterized by tissue hypoxia, as methemoglobin cannot bind oxygen. (1, 5)
Carcinogenicity (IARC Classification)Ingested nitrate or nitrite under conditions that result in endogenous nitrosation is probably carcinogenic to humans (Group 2A). (2)
Minimum Risk LevelNot Available
SymptomsNitrate and nitrite poisoning causes methemoglobinemia. Symptoms include cyanosis, cardiac dysrhythmias and circulatory failure, and progressive central nervous system (CNS) effects. CNS effects can range from mild dizziness and lethargy to coma and convulsions. (4)
TreatmentMethemoglobinemia can be treated with supplemental oxygen and methylene blue 1% solution administered intravenously slowly over five minutes followed by IV flush with normal saline. Methylene blue restores the iron in hemoglobin to its normal (reduced) oxygen-carrying state. (5)
