ClassSmall Molecule
Description
Azathioprine is an exogenous solid with the formula C9H7N7O2S and an average molecular weight of 277.26 g/mol. Azathioprine belongs to the aryl thioethers, a subclass of thioethers within the organic compounds. It is found in or released from healthcare, pharmaceuticals and veterinary products used as immunosuppressants, as well as food, food processing and cookware used for food preservation. Recorded exposure routes include oral and intravenous administration. As an immunosuppressive pro-drug, azathioprine is converted into 6-mercaptopurine in the body, where it blocks purine metabolism and DNA synthesis. It antagonizes purine metabolism and may inhibit the synthesis of proteins, RNA, and DNA, while interfering with cellular metabolism and inhibiting mitosis. Its mechanism of action likely involves the incorporation of thiopurine analogues into the DNA structure, resulting in cytotoxicity and chain termination. Recorded protein targets include Ras-related C3 botulinum toxin substrate 1 (RAC1), Amidophosphoribosyltransferase (PPAT), and Hypoxanthine-guanine phosphoribosyltransferase (HPRT1), of which it is an inhibitor or modulator.

Mechanism of ToxicityAzathioprine antagonizes purine metabolism and may inhibit synthesis of DNA, RNA, and proteins. It may also interfere with cellular metabolism and inhibit mitosis. Its mechanism of action is likely due to incorporation of thiopurine analogues into the DNA structure, causing chain termination and cytotoxicity. The oral LD50 for single doses of azathioprine in mice and rats are 2500 mg/kg and 400 mg/kg, respectively.

Symptoms may include feeling tired, shortness of breath, easy bruising and bleeding, and increased risk of infection. Occasionally, spread may occur to the brain, skin, or gums. As an acute leukemia, AML progresses rapidly and is typically fatal within weeks or months if left untreated. Risk factors include smoking, previous chemotherapy or radiation therapy, myelodysplastic syndrome, and exposure to the chemical benzene. AML affected about one million people and resulted in 147,000 deaths globally. It most commonly occurs in older adults. Males are affected more often than females.PMC11130641

myasthenia gravisNot AvailableNot AvailableMyasthenia gravis is a chronic autoimmune, neuromuscular disease that causes weakness in the skeletal muscles that worsens after periods of activity and improves after periods of rest. These muscles are responsible for functions involving breathing and moving parts of the body, including the arms and legs. The name myasthenia gravis, which is Latin and Greek in origin, means “grave, or serious, muscle weakness. The hallmark of myasthenia gravis is muscle weakness that worsens after periods of activity and improves after periods of rest. Certain muscles such as those that control eye and eyelid movement, facial expression, chewing, talking, and swallowing are often (but not always) involved in the disorder. In myasthenia gravis, antibodies (immune proteins produced by the body’s immune system) block, alter, or destroy the receptors for acetylcholine at the neuromuscular junction, which prevents the muscle from contracting. This is most often caused by antibodies to the acetylcholine receptor itself, but antibodies to other proteins, such as MuSK (Muscle-Specific Kinase) protein, also can impair transmission at the neuromuscular junction. Myasthenia gravis affects both men and women and occurs across all racial and ethnic groups. It most commonly impacts young adult women (under 40) and older men (over 60), but it can occur at any age, including childhood. It has a prevalence of about 1 in 5000 individuals.PMC1492602
autoimmune diseaseNot AvailableNot AvailableAn autoimmune disease is a condition arising from an abnormal immune response to a functioning body part. There are at least 80 types of autoimmune diseases. Nearly any body part can be involved. Common symptoms include low grade fever and feeling tired. Some autoimmune diseases such as lupus run in families, and certain cases may be triggered by infections or other environmental factors. Some common diseases that are generally considered autoimmune include celiac disease, diabetes mellitus type 1, Graves' disease, inflammatory bowel disease, multiple sclerosis, psoriasis, rheumatoid arthritis, and systemic lupus erythematosus. About 24 million (7%) people in the United States are affected by an autoimmune disease. Women are more commonly affected than men.PMC10669396 , PMC3994740
